I would think to be called a major city, it needs to have at least 100,000 people....

Well, the criteria, according to me, is that either it has a large population, as y'all stated, or is a significant place along the highway. For example, the Texan city with 23,000 people---even though that's relatively small---has the intersection of I-35 with another interstate highway. Or take the small town in Kansas with 24,000 people. It's the place where a spur forms from I-35, which is significant to me.
